Curcumin is a potent inducer of the heat shock response with complete interference of the heat shock gene Sirtuin 1 (Sirt 1) regulation of heat shock protein 70 (HSP 70). Sirt 1 is the heat shock gene with its repression associated with insulin resistance and programmed cell death. Curcumin at higher doses may override Sirt 1 activators/modulators with induction of mitophagy and NAFLD.

Recent reports for the use of Curcumin have indicated that this polyphenol has not yet been approved for human use. Curcumin and its poor bioavailability and limited adverse effects reported by some investigators are a major limitation to the therapeutic utility of curcumin. The authors warn of expected results from ongoing clinical trials that may provide a deeper understanding of curcumin’s therapeutic potential and will help to place this fascinating molecule at the fore front of novel therapeutics.

The effect of curcumin on the nuclear receptor Sirtuin 1 should be veiwed with caution since high doses in human cell studies shows that it interferes with the cell’s heat shock response. Curcumin and its effects on heat shock proteins (heat shock protein 70) should be carefully assessed since HSP70 has been associated with programmed cell death. The heat shock gene Sirtuin 1 is critically involved with HSP70 regulation and Sirtuin 1 regulation by Curcumin/turmeric needs to be assessed with higher doses that may inactivate HSP70 metabolism in various cells and tissues in man.
