Blood volume and the serum obtained from the same amount of blood are different but the glucose content of that much blood is same.
Is there any method or standard, defined ......?
Glucose is estimated with handheld meters directly from blood and it is also estimated by enzymatic methods available as kits.
So what is the standard ?

We usually say serum glucose when we used your mentioned methods
Sometimes we used glucometer for capillary blood ie. whole blood

Earlier blood glucose was measured in whole blood. Nowadays serum is extracted from blood and glucose is measured in the serum. Whole blood and serum blood glucose is often different. Red blood cells have higher concentration of protein than serum and serum has higher water content and more dissolved glucose than whole blood. To obtain blood glucose in serum from figures in whole blood, it is multiplied by 1.15.

The answer is "NO". Once we talk about BLOOD GLUCOSE, we intend whole blood which is usually measured by glucometer or some other point of care instruments. What is has is red blood cells which only consume glucose as diet so levels if not immediately check can cause falsely low results. On the other hand whole blood is affected by blood cell content and water/serum content. Hemoconcentration would lead to a high glucose result and vice versa. More so it is not recommended by FDA for diagnosis of diabetes mellitus. Contrary to that plasma or serum glucose levels are not affected by red blood cells or the factor haemoconcentration or dilution and is recommended to be used for diagnosis of diabetes mellitus. Normally, labs do these testing and is usually considered better than blood glucose method.
