I am master student in parasitology Iran. Actually In my thesis I checked the isoenzyme system of Fasiloa hepatica and gigantic by means of Tetrazolium staining. As you know in this method we can see some violet band. Surprisingly, in my experiments I observed some white bands in addition to violet band and I couldn’t find any reason for these results.
Is anyone here who knows what the reason for getting these additional bands in my staining? I attached a picture from one of my experiments.

Hi Bahare, I don't have any experience with your assay system, but the color of tetrazolium salts depends on their reduction to formazan compounds.
Their color can be dependent on the presence dehydrogenases and reductases.
My guess is that the clear bands are enzymes that inhibit tetrazolium reduction.
Most likely, the white bands primarily reflect superoxide dismutases (SODs).
The main tetrazolium-reducing compound is superoxide radical. This reaction is inhibited by SODs. In fact, we once used this to show superoxide production in mucus. http://www.iovs.org/content/16/8/762.long
