Hi, I am not an expert in cell line. I think you can simply transfect cell lines like HEK with plasmid containing NMDAR gene by electroporation or virus. In my limited knowledge, lentivirus will link your gene into the 4th or 6th chromosome of the genome. Therefore you can expect stable expression for your cell line. I may check which chromosome it is if you need.
Thanks Yu, much appreciated ! The problem with NMDAR is that they become toxic for the cells so it's a bit tricky in that manner as compared to expressing a protein that won't harm the cell.
If you keep your external Mg at 2-3 mM and perhaps reduce Ca to 1 mM, and if you maintain your cells healthy (membrane potentials more negative than -60 mV), NMDA receptors should just quitely (inactive) sit on the membrane waiting for your commands.
Alternatively you may think of expressing your NMDARs in lipid bilayers.
