I chose this wavelength as the sterile glucose medium that I use doesn't provide any optical density at this wavelength but only when E coli is growing in it. I've also correlated optical density values with accurate E. coli count (at this wavelength) and I found a good correlation value (r=0,93). But it seems that this wavelength has been seldom used for E. coli. Can anyone suggest why?
This means that there is regeneration through substrate deposition , please carry out other test for micro-elements.
The normal wavelength for measuring bacterial density is 600 nm, although 680 should be fine. It is important to realize though that you are not actually measuring light absorption in this assay, but rather light scattering. The more bacteria, the more light scatter and the less light hits the detector, so you are not really measuring the absorbance of anything. If it works, go for it.
