For an experiment want to try, i need to grow my iPSCs in suspension. The cells i would use, have been grown as adhesive monolayer on vitronectin, in mTeSR E8 medium until now.
I already tried a couple of things: seeding them in low attachment u-shaped plates and in agarose coated wells (from a publication) but couldn't obtain the aggregate i was hoping/expecting to get.
Anyone experienced in culturing iPSCs in suspension as aggregates? Any useful tips? Thanks a lot!
I believe it is not possible, unless you treat them with CHIR, which will lead differentiation... but 2023, maybe you made it :)
Blerta Stringa in the end I did manage to obtain iPSCs aggregates by culturing in low adherence culture dishes (96-wells plates, u-bottom shaped wells) by adding poly-vinylalcohol to the medium.
