Ola, that would be tricky for the obvious reason. ;)
If you mean secretion of functional insulin, that would be a different matter. In that case, you should do a literature review, for example (by no means the only one or the most useful one!):

You would find that many investigators have put in a considerable effort in a variety of immortalized insulin-secreting cells for well over 2 decades, optimizing and tweaking insulin secretion profile to mimic the native beta cells. I do not expect that anyone can provide you a straight-forward answer that if you do x, y, and z, you will have what you need.
