I'm working to isolate nuclei from human liver tissues, for single-nuclei RNA seq purposes. While I was able extract nuclei, there was still a lot of liver cells that were not lysed. I've attached an example image that was taken at 20X resolution.

Have you encountered something similar?

Lysis buffer tested:

Invent SN-047 lysis buffer

Hypotonic nuclei lysis buffer (20mM Tris-HCl, 10mM NaCl, 3mM MgCl2)

We have also tried using a Dounce, and adding NP40/Tween-20 into the lysis buffers, to no avail.
