The nanoparticle remains dispersed in water, but when treated with equal volumes of DPPH in methanol. The nanoparticle may become solubilized in methanol and the drug may come out of its lipid core, in that case how can it be guaranteed that the antioxidant activity is due to the nanoparticle not the pure drug?
Is there any other alternate assay in specific for lipid type nanoparticles?

You can do a release profile and determinate the drug concentration vs time .. Can use this information for know the max concetratiõn and take the aliquots ("eluate" ) in your experiment with DPPH.
