Hi everyone. I isolated a lytic phage for wound infection caused by S. aureus. It didn't make a clear spot on the lawn of ATCC 25923. Also, I examined it on 24hr formed biofilm of this bacteria by tissue culture method. in comparison with SM buffer as a negative control. data were collected from 6 repeats and analyzed by post-hoc (p value= 0.005).
Does anybody have any clue about biofilm increase in presence of a lytic phage?

I have struggled to understand your question, but the final sentence I interpret as meaning your bacteria seemed to produce more biofilm when the bacteriophage was present. It could indicate that infected bacteria release some metabolite and this serves as a warning to nearby bacteria. These bacteria then put more energy into producing the secretions which produce the biofilm. This would, or could, help prevent the phage from being able to make contact with neighboring bacteria and causing infection.

The method of biofilm evaluation in the article is different from my method. also, I couldn't find how can I check the metabolic growth cycle in biofilms.
