I want to do the Immunophenotyping of the Human Dendritic Cells markers (CD80, 86, 83, HLA-DR etc) after stimulation with TNFa & oxLDL. At the moment FACS is not available in my department. So can I proceed this with real time PCR ?
You can do Immunophenotyping with RT-PCR but the result will not be comparable with Flow cytometry. Because the only one or two marker positive cells cause the positive result but in flow cytometry you can see the number of marker positive cells with frequency of marker on cells.
You can use a PCR array for the evaluation of the genes expressed in your population of DCs. This approach is interesting due to the fact that you can see lots of different genes in isolated, retrotranscribed and amplified mRNA, which will be ultimately translated to proteins (most of them, think about post-transcriptional regulation of gene expression).
On the other hand, as previously cited, the results are not comparable to flow.
