I have performed anti-Oct-4 staining with A488 on a colony of hiPSCs. In ZEN, I have applied an intensity channel (rainbow) to the image. As you can see, the highest intensity is located in the nucleus of the cells.
Is there any way to add an intensity scale bar to the image?

Calibration bar cannot be applied to RGB or Composite image, You need to first reduce the colour depth then apply gradient to the image before inserting the Calibration bar.
Try the following steps:
1. Convert the Image to 32-bit; Image>Type>32-bit
2. Apply LUT; Image>Lookup Tables>Viridis (or any 3-level colouring gradient is suggested although 2-level gradient will also work fine)
3. Insert Calibration bar; Analyze>Tool>Calibration bar.
