I want to know if someone can give me the optimized concentration of anti-mouse IL-5 used in an ELISA test and maybe advise me about which clones I should use.
I'm currently working with clone TRFK4 (biotin) and TRFK5 (purified) and I didn't manage to see even my standard curve (by using maximum concentration given in TDS). No detection of cytokines.
I hope someone can help me.

Hello,
Finally, the problem was because of my standard cytokines. It seems that a degradation happens during the conservation at -80°C, in a RPMI medium. A newly prepared standard worked with my conditions and those clones.
