The best way should be to use of a special hypoxia chamber in which you could maintain very low concentration of oxygen. If not, you could make an artificial hypoxia by adding 100-200 uM of fresh CoCl2 solution into cell culture medium. Just it is important to replace the medium by fresh CoCl2 solution every 2-3 days.
If your lab has the resources the best method is to use a glove box hypoxia chamber.
If not a MIC-101 hypoxia chamber is also widely accepted. You can flood the chamber with low oxygen and incubate for the desired treatment time.
Most treatments range from around 6h -48h at 0.5-2% oxygen concentrations.
Chemical inhibitors such as cobalt chloride and DMOG can also be used; however you cannot be sure if they target other pathways.
