I washed by QC buffer and I put the eluted buffer and forget to remove the washing buffer from the tube? Now I have tube having washing buffer and eluted with DNA. How can I solve this problem?
I would pass all the flow through from the column again. The QC buffer will pass and some of your plasmid will stick in the silica membrane again. Discard and spin again to dry your membrane and then add your elution buffer. Probably you will recover 60-70% of your plasmid.
The yield of your elution is then not optimal as the remaining washing buffer tends to let DNA adsorbed to the column. What I would do is to repeat elution from the column to get the rest of the DNA eluted and then compare what is in the first and second elutions... If you want to recover the DNA from the first elution into elution buffer it's either repeating column purification or through DNA precipitation.
