Hey! I need help on peptide (22 aa) oxidation.This peptide has no methionine, 1 tryptophan & 1 tyrosine. also has 4 cysteine so I used 2 protection groups (trt & acm). For ACM removal, I am using 0.1 M iodine solution in 0.1 mM peptide followed by 3 minutes N2 blanket and dropwise ascorbate (1 M) addition.

I’m getting an extra mass of 32 dalton in HPLC & LCMS peaks Which probably is due to Tryptophan oxidation.

It would be of great help if I may know how to get rid of it.

the first peak is (2586 dalton) & the second peak is (2754 dalton) as shown in the pictur.
