OPM2 cells grow rather slowly and are more sensitive among others. But, I am unable to get a good percent of viability when I passage these cells. Most of the times, the cell death is much higher than expected.
I have been using RPMI media with 20% serum. Any guidance or suggestions? Has anyone else faced such issue?
Assuming that you received the original cell vial from a reliable source (e.g., ATCC) and you have maintained the cell culture as noted in the cell supplier's description (see link below), it is likely that cells have undergone genetic changes, thus, altering their cell survival requirements. If cells are maintained at appropriate cell density (up to million cells/ml) and still exhibit poor or less than optimum viability, it may be better to get an original cell vial from a reliable source for proposed studies (instead of selecting a cell population that survives your culture conditions).
