I am working on toxin production responsible for gas gangrine by using clostridium perfringens in static flask condition but i will able to get only small amount of toxin by this method. Can any one suggest me how to scale up to get higher yield of toxin with good toxicity?
Hi first of all I haven't worked on Clostridium. One suggestion is that you can enhance your specific medium with nutrients which will aid in production of large quantities of the toxin. I had a similar problem while working with P. aeruginosa so I enhanced the specific media which I used with other nutrients. For this you may have to do a careful study on the pathways and trial and error methodology on the media composition will yield fruitful results.
A question here, do you face very less activity of the protein without the scale up, then you may need to check your ptrn purification process. May be its coming down harsh on your protein to lose activity.
