I am running 30 ug-40ug proteins and looking at several proteins ranging from 85kDa to 25 kDa. We have been using a wet transfer system with a good assessed transfer by amido black staining. The primary incubation is over night (1:5000 to 1:10, 000) and the secondary is for 1-2 hours at RT usually at 1:10,000 but sometime increasing to 1:2000. We have tried Clarity (not so good), our old Perkin Elmer was ok for one protein, and GE Health for actin (not the best ever though it should be very strong for 1:10,000 overnight incubation (Santa Cruz HRP conj). Sometimes we get good bands and sometimes not. The other lab uses BioRad Immunostar, but I have been told by BioRad that BioRad will not longer make this, and they suggested we order Clarity. We do the ECL incubation on the bench at RT for 1-5 mins depending on the reagent recommendation.
