You're saying that you get a signal when the Bicore loads a blank sample, even though the blank sample is actually the same composition as the running buffer. That means you're getting a contaminant from the sample loading valve or loop. Try flushing it by injecting neat DMSO a few times, or 8M guanidine, or 1% SDS, or 70% IPA.
