I am getting a thin white layer in my lymphocyte culture. I used dmem media with 10% fbs to culture lymphocytes. After culturing 24 hours in CO2 incubator, i observed a thin white layer formation at the base of the well. can someone plz tell me if it’s the cell monolayer or Something else. I’m attaching pictures.

I am not very sure, but I would still like to suggest.
When you transfer the PBMC layer to a 15 mL conical tube, do you wash the PBMC with PBS ?
After you separate the PBMC layer, you should wash the PBMC twice with PBS, centrifuge cells at 500 x g for 5 minutes each time. You will notice that the supernatant will be somewhat cloudy after each wash. Multiple washes in PBS is necessary to remove any residual Ficoll.
