Yeah, sufficient amount as well as the purity is good. I am doing sequencing in house.

reaction conditions may not be good for primer annealing. Whats your temp and buffer?

Annealing temp. is 50 degree Celsius, and we used regular sequencing buffer from ABI.

can you post an .ABI file of the failed sequencing please.They contain a lot of useful information. Are all of your sequences failing and have you checked that the primer sequences exist in your vector. Are you sure that your sequence is in the vector on the samples that you are sequencing
