The fungus i am using is Mucor hiemalis and i am doing layer by layer coating using chitosan and pectin on LDPE film. I want to see antifungal activity (inhibition zone) of the coated film. The concentration i used for chitosan is 0.6%, 1.40% and 2.2%. I used disc diffusion method for this purpose where i use coated film as disc. But i am facing problem as it is not showing inhibition zone.

The chitosan concentration seems to be correct and it showed antifungal activities in some reported works. How long (days) the experiment was carried out? I recommend conducting tests for longer days (For eg., 9 or 12 days) and use both positive and negative controls to study the pattern of fungal growth.
