I am currently differentiating 3T3-L1 cells and I am uncertain about the presence of lipid droplets. However, before conducting the Oil Red O staining, I would like to know if there are already lipid droplets present. Can someone provide me with this information?

Usually, lipid droplets should appear within 4-7 days after differentiation is induced. They will look extremely small initially. Lipid accumulation continues throughout the first two weeks. The lipid droplets gradually fuse to several big locules.

The figure you attached is which day after you induced differentiation. I am still in doubt whether the arrows in the fig indicate lipid droplets.

From the picture it does not seem there is much lipid accumulation in that microscopic field. Lipid droplets in 3T3L1 look a bit different. You will see multiple droplets inside a cell within 4-7 days.

You didn't mention how many days the culture in differentiation, but it seems like the differentiation process has begun. The cells are starting to change shape, but you still can't see any lipid droplets
