I conduct pressure myography experiments on mesenteric arteries. The physiological saline solution is bicarbonate balanced and pH with 5% CO2, 20% O2 and nitrogen balanced, back temperature is measured at 37oC. Solutions are made up fresh the same day of experiment, same goes for hydrogen peroxide. H2o2 is not added to solution until moment before changing solutions. I know from my previous experiments and literature that h2o2 should (10^-7, -6 and -5 M) should induced an increase in vessel diameter. However, my experiments the vessels are constricting or not dilating at all. HELP!?
