We are using HUVECs to study the mechanism of kinins in inhibitingn TNF-a-induced expression ICAM and P-selectin by endothelial cells, but we kept finding big variations in the results: sometimes bradykinin potently inhibited the effects and sometimes the inhibitory effect is absent. We tired to use cells at passage 3, 4, 5, 6, or 9 and we still see variations. Some peers suggested the use of cell line of endothelial cells. Could anyone help us out with the issue of selection of endothelial cells and whether to passage a primary cells (if yes how many passagess) or use a cell line (any suggestion)?
