How to use NCBI BLAST to obtain proteins from nucleotide sequence?

I am screening for AMR genes and heavy metal resistant in S. aureus. Would it be recommended to use NCBI blastx to obtain these genes?

What parameters would be recommended to use on NCBI BLAST to yield the best results?

My rosetta strain has chloramp resistance and i am transforming my plasmid with amp resistance but not getting colonies on chloramp/amp LB plates?
