I've been using cryopreserved PBMCs from ATCC and iQ Bioscience and found it weird that in their manual, they have direction for thawing the cells but do not have re-freezing the cells.
My colleague told me to aliquot the vial with about 10-50MM cells into smaller cryopreserved vial upon arrival and re-freeze them for later use, but I've noticed a sharp drop in cell viability.
Is it normal to thaw and re-freeze the cryopreserved PBMCs?
Best will be thaw PMSCs, wash away freezing medium, place in medium for PMSCs for some hours to recover membranes, than concentrate PMSCs and place in 2x fresh freezing medium and than aliquot in vials.
In my experience refreezing PBMCs is close to impossible. The thawing alone already stresses them a lot und if you refreeze the immediately you will likely loose all of them. You could try keeping them in culture for 1-2 days but you will get a selection and not the whole/ identical composition.
They are best used fresh.
I don't recommend re-freezing cells, your viability will take a big hit. Cryopreserved monocytes have major problems with antigen presentation.
