I want to somehow label my protein surface. I wonder if I can modify my protein by using some specific enzyme i.e. post-translational enzymes. Any enzyme is OK. Is there any common and economic way to do it? Of course, I don't expect too much effect on structural conformation. I can detect the structure by using NMR which is not a problem for me.
Any enzyme, any reaction or any idea is welcome.

Hi Qian Wang You don't need enzymes. If you want to label the protein surface it would help to know what amino acids (aa) are exposed on that surface.
There are simple and cheap chemistries available to modify the most abundant aa functional groups. The most common protein functional groups are primary amines (-NH2) and carboxyl groups (-COOH).
You can pick and choose what you want to attach to these groups.

which is the aim of your esperimenti? And what do yuo mean for uniform? it means all the residues exposed in the surface? because the most labelling approaches are targeting specific aa ah lysine or cysteines and therefore is a selective and not uniform lanelling. also glycosilation happen in specific residues as N , S or T. if you would like your protein surface by nmr a possibile stategy is perform hydrogen/deuterium exchange where you will see ti disappear all the signal related to exchangable surface exposed deuterium.

Manuele Martinelli Sorry for the confusion. I want to cover as much area of the surface of my protein in order to find the interacting surface. Yes, I am trying the HD exchange. I am just thinking of another backup plan for identifying the surface. Probably I should try a combo of various reaction to label the aa.
