how do i prepare the various concentrations of an emulsion for drug treatment? i have prepared an oil in water emulsion encapsulating docetaxel. to treat my cells (mcf-7). I need to prepare serial dilutions and then perform mtt assay to obtain the ic50 value. how do i go about this? do i extract the docetaxel in a suitable solvent and then treat my cells? or do i use the emulsion as is? but how to prevent turbidity when adding in serum free dmem media?
Emulsion turbidity (serial dilutions) could be measured prior to the MTT assay and the values subtracted from the assay measurements. However, it may not be optimal for the result, because it may mask the cell viability measurements.
not sure why you would perform measurements in demineralised water? Would this not affect your cells?
Titus Sobisch so when i add my emulsion to serum free media it seperates but i dont have an alternate to media.. how else can i go about this issue?
