If you have your CD14+ cells isolated (monocytes) you can differentiate them into macrophages by incubating them in RPMI 10% FBS supplemented with M-CSF (50ng/ml) during 5 days.
depends on which macrophage profiling you need. MCSF to start with but if you are looking for M1 phenotype i recomend IFNgamma and LPS, for M2 il4 and il10
To isolate monocytes, you can either use CD14 microbeads (Miltenyi or Biolegend but I prefer Biolegend) or simply isolate PBMCs and plate them in polystyrene 6 well dishes at 37 degrees CO2 incubator for 2 hours and aspirate the medium.
For macrophage differentiation, depending on your choice of polarization, for M1 use GM-CSF + IFN-g or LPS; for M2, use M-CSF + IL-4-IL10/TGF-b. For M0 do nothing. Culture them for 7 days and viola you have your macrophages.
