I'm looking for assays where I can track single cell - single bacterium dynamics, mainly rate of killing. Most methods I find online revolve around finding a single infected cell and following it via live cell microscopy. Are there any other methods, where I can follow the course of infection in a single cell, such that I can measure bacterial killing/survival in macrophages? Thanks!
Why does this need to be at the single bacterium level? For Listeria, we infect macrophages, kill remaining extracellular listeria with gentamicin, lyse the macrophages with dH₂O some time later, plate onto blood agar plates, and count listeria colonies to quantify the number of live listeria in the macrophages at the time point.
For such a high end single cell analysis, do you have such a high end platform i.e. microfluidics or high resolution/confocal microscope?
