Null Hypothesis (H0): Elevation has no significant effect on soil carbon content when soil depth is not considered as a variable.
Alternative Hypothesis (Ha): Elevation has a significant effect on soil carbon content independently of soil depth. One of the means to test this hypothesis that i can imagine is conducting a series of one-way ANOVAs with subgroup analysis within specific soil depth ranges.
There needs to be a clarification about what you mean when you say "elevation"? Do you mean elevation above sea level or some other common baseline or do you mean elevation within a landscape which could be considered as landscape position.These are potentially two very different questions.

This would be an interesting study. When studying SOM and elevation one must also consider other topographic features. such as aspect, relief and slope. These factors could impact SOM through erosion processes, for example higher erosion on slopes and deposit at the toe of the slope. Elevation may also impact soil temperatures and potentially rainfall and vegetation, all have the potential to influence SOM. These are likely only a few factors that would influence SOM.
