There're four kinds of virus genome in a virome sample: ds/ssDNA and ds/ssRNA. we want to use a viral DNA/RNA kit to purify the mixed genome and sequence it in a run via pyrosequencing.
If we first use reverse transcriptase on the ds and ssRNA to synthesize the first cDNA strand, then add the adapters before sequencing by ion-Torrent or HiSeq, can the dsDNA and first cDNA strand be sequenced efficiently?
Do I have to sequence the RNA and DNA genomes separately?
