Alternately, you can obtain blood via retro-orbital bleed. Be sure to use heparinized or EDTA-coated microcapillary tubes [50 microliter] to collect the blood. This is essential to prevent clotting and allow you to examine the blood cells if you are using flow cytometry to examine the leukocytes. Alternately, if you want to do a blood smear, use untreated microcapillary tubes, then place a small drop of blood on a microscope slide and use a 2nd slide to make the smear. Air dry, fix with methanol, stain with Wright Geimsa and examine under hi-dry conditions.
also I Wish to inform that once isolated BM cells i.e. the lin- cells. I FACS sorted them and did CFU assay with the sorted cells. will you advice to perform lentiviral transduction on the CFU plate or I grow cells in liquid culture. Can anyone suggest or advice
