Hi Ratish, tail bleeds can be used for anything blood related.
In one of my previous jobs, an inept animal handler mixed together several gene knock-out (KO) mice when changing their bedding and cages.
To reisolate the correct KO mice, a technician did tail bleeds of all 120 mice and determined by PCR the KO and reestablished the correct colonies.
Alternately, you can obtain blood via retro-orbital bleed. Be sure to use heparinized or EDTA-coated microcapillary tubes [50 microliter] to collect the blood. This is essential to prevent clotting and allow you to examine the blood cells if you are using flow cytometry to examine the leukocytes. Alternately, if you want to do a blood smear, use untreated microcapillary tubes, then place a small drop of blood on a microscope slide and use a 2nd slide to make the smear. Air dry, fix with methanol, stain with Wright Geimsa and examine under hi-dry conditions.
Hi Ratish, blood is blood no matter how you collect it.
If the information you need is in blood, then it you have to devise the best way to collect a sample.
Tail-bleed, retro-orbital bleed, hind-leg bleed, etc, doesn't matter as long as you get the information you are looking for.
also I Wish to inform that once isolated BM cells i.e. the lin- cells. I FACS sorted them and did CFU assay with the sorted cells. will you advice to perform lentiviral transduction on the CFU plate or I grow cells in liquid culture. Can anyone suggest or advice
