I am isolating vibrios from a marine source, a few of my strains are not able to grow again during subculture. Can anyone suggest methods to revive them?
what media are you using? I usually use marine agar/broth for marine bacteria (including Vibrio diazotrophicus) http://www.bd.com/europe/regulatory/Assets/IFU/Difco_BBL/214907.pdf
I also use marine agar to isolate marine bacteria and yeasts but in some cases I have to adjust the concentration of salt in the medium for better isolation
Some heterotrophic micro-organism growth better in poor nutrient agars. Like R2Agar for drinking waters. With long times and lower temperatures...maybe in this case you'll need to increase salt concentration.
Try co-culture with amoeba from genus Acanthamoeba. Many bacteria, including vibrio can resuscitate after growth inside amoeba cells. This method is also used for bacteria hard to cultivate on popular microbiologic media.
