I am using the EZ-Link™ Sulfo-NHS-Biotinylation Kit from Thermo Fisher in order to get biotinylated antibodies. The kit seems to work well, but I have problems when removing the excess of unbound biotin. I have tried using the Zeba-Spin desalting columns from the same kit, using a centrifugal filter with polyninylpyrrolidone (in order to avoid the antibody from sticking to the membrane) and also throught dialysis (2 dyalisis: one ON and the other for 5h), getting only to recover the 25% of the antibody after these steps (in all cases). Any ideas about how I can recover higher amount of antibody? or another methods to do it?
