I need a method to isolate mosquito cell membrane protein from whole mosquito abdomen. Therefore, I will need to remove the mosquito hemolymph and exoskeleton.
I have not find a clear published method to do that. Any suggestions would be super helpful

Hi, there is no way to remove the tegument chemically; you need to dissect and open the abdomen, in order to recover the internal organs and tissues. You can simply cut the tegument along the abdomen, and given that you do not need to conserve the integrity of internal organs, you can scrap the interior with a needle. If you are not habituated to dissecting insects, you would probably need to practice a little bit before having a proper result, but it is not difficult. I recommend using a micro-scalpel made of pieces of a breakable razor blade and Ringer solution for avoiding the tissue to get dry.
