Blood from some donors are fatty, after centrifugation an upper layer forms which is difficult to remove, in order to get clean plasma for cfDNA analysis.
Any advice? Thanks!
Hi Mariana,
Hope everything is going well.
You could use a organic solvent. Here I attach a publication talking about that.
regards,
Domingo
maybe you could try piercable centrifugation tubes and simply remove your plasma-aliquot with a Syringe from beneath the fat-layer.
Best regards,
Christian
Yes Dr Christian Wechselberger fat will separate by density gradient also
Thanks to all!
If you put the centrifuged plasma in a -20 freezer for 10 or so minutes the fat will solidify and you can then remove the plasma from underneath.
We are receiving blood samples that sometimes we can't make to stain them freshly for FACS. Is it better to keep them ON in RPMI+FBS10%, cold PBS (4ºC), or cold methanol? We perform both...
