I am working on one project for Assay and relative for PEG (MW about 20000Da) by HPLC-ELSD at 1.5% (w/w) level. However, the relative high concentration of Na Hyaluronate (3% in w/w) with high viscosity causes the in-accurate injection volume withdraw. Is anyone here having any suggestion to reduce the aqueous sample viscosity without further dilution?

You can always change the withdrawal velocity of the needle. I don't know if you can use hyaluronate of reduced molecular weight or to heat up the sample.
Good Afternoon, Omar. I try to varying the withdrawal velocity of the needle, but it does not help, as the high viscosity of Na hyaluronate. But I do try to heat up the samples, and it seems the issue get improved. Thanks again.
In general, increasing the ionic strength will reduce the viscosity of a polyelectrolyte solution because this shields the charges along the polymer backbone. However, your ELSD will probably not like most salts. Volatile salts salts as ammonium formate or ammonium bicarbonate might be an option. You may also try to decrease the ionic charge along the polymer backbone by decreasing the pH for example with TFA (0.1%).
