I am trying to quantify different viral proteins by running ultracentrifuged virus on SDS PAGE and visualizing the gel on a gel doc. Is there any other method to quantify individual viral proteins apart from this?
Different viral proteins can be detected by its specific antibody or a serum that is raised against the virus(to detect all in one)
It is better to measure the protein level before SDS-PAGE by simple method such as Bradford.
However, after running, you can use densitometer for quantifying of the protein bands.
Another simple and rapid method also existed. This is absorbance method.
If you can get good staining on SDS-PAGE, then that seems like the easiest method. Remember to correct for the different sizes of the proteins when quantifying staining. You could also separate the proteins by chromatography and use UV absorbance to quantify them, taking into account their amino acid compositions.
