Is there anybody out there who has experience how to isolate and purify common inflammatory cytokines (TNFa, IL-1b, IL-6 etc.) from inflamed mouse ear tissue? We usually do it with blender using phosphate buffered saline (PBS) with protease inhibitors (PMSF) on ice, and it works very well with many tissue types, but unfortunatelly we got very poor results with ear tissue when measuring cytokines with Luminex assay.
