I've seeded my hMSC cells in T25 flasks (in 5 ml medium), the cells will be incubated for 5 days. After 5 days, I will use the Biocolor Sircol assay kit to determine the collagen I production from my samples. The kit recommends using cold pepsin-acetic acid to harvest the collagen from ECM after removing the spent medium. However, there is no detailed method on how to exactly do it.

So, after removing the spent medium, do I need to trypsinize the cells or just scrape and collect everything from the flask surface and then add in the pepsin solution. Anyone can help me with this? Thank you.
