I'm working with rabbit tibial defect model. After the completion of follow up time period, I'm gonna extract tibia from the animal and store for an undefined period of time. The bone sample will be processed for micro-CT analysis, mechanical testing and histology. However, I'm not sure about the storage conditions of bone tissue. Can storage in 10% formalin affect the mechanical properties of the bone, as the procedure I'm following for Micro-CT and mechanical testing do not ask for fixation of the bone tissue?
I just need to know the temperature and solution in which the intact bone can be stored..

For the histology I don't recommend keeping them in the formalin. Fix the bones then store in sterile PBS at 4oC. Remove and replace the PBS weekly and the bones will be fine.
PBS (10x) e freezer com menos de 70 graus Celsios.
Trabalho com isso a muito tempo. Nessa temperatura com o PBS pode conservar a vida toda.
Marina, I have just one query. PBS will freeze at -70°C, so how is it supposed to preserve the bone sample?
Aditi, eu trabalho com tecido ósseo a 8 anos e as minhas amostras são cogeladas nessa temperatura e nunca perdi nada. Não troco o PBS regularmente, apenas após o sacrificio congelo em PBS a -70 ° C, e assim minhas amostras são todas preservadas para o projeto em questão.
Acho que não entendeu......Entendi o que vc queria saber. O PBS mesmo congelado ele conserva a amostra óssea. Sim, isso é baseado em artigos. Posso até enviar as referencias.

We (Bryan Wright 1st author, Biomechanical lab., Orthopedic departmenet, Oslo Univeristy Hospital) have just submittet a manuscript where we find intact mechanical parameters compared with qCT parameters and conclude that formalin does not seem to interfere with biomechanical parameters.
