Can anyone tell me how prepare chitosan nanoparticle ( preparation with ionic gelatin) without agglomeration after freeze drying or centrifugation. Because agglomerated particles have not measurable size with TEM and SEM. I used 5:1 (CHITOSAN:TPP) for synthesis and tried evading agglomeration using 0.05% CTAB.
You can use small molecules with hydroxyl groups like trehalose, dextrose.
Basically you can't. Once dried you have a collection of fused sub- and post-micron aggregates with no discrete, free, indepedent particles of < 100 nm. See:

You can sterically stabilize the system by forming the nanoparticles on an (inert?) substrate such as found in the catalyst industry (e.g. 5% Pd on C).
I used trehalose before freeze drying the chitosan nanoparticles. But as Dr. Rawle explains dried particles are greater than nanometer size when measured by DLS.
Actually Dr. Swati, when particles are dried two naked particles have the opportunity to come closer and they started agglomerating resulted bigger/mega size particles. So, there is every possibility to be greater than nano-meter size.
