How to prepare rice leaf sample for confocal image without affecting its cytosolic and organal pH?

Dear Friends,

I want to analyse my rice shoot and root tissue for confocal image analysis to look its pH changes through the ratio of emission wavelength intensity of acridine orange dye.

I have got some article regarding fixation and dehydration of tissue but this process might change the pH of the plant sample.

In some other articles people have mentioned directly hand trimming method to observe the cytosolic content, regarding that I am bit doubtful would it be helpful to analyse the sample or not.

My target is to observe vacuolar and cytosole pH changes during vegetative growth period of rice seed ( 5 days after germination).
