I would like to work with organotypic culture of liver slices, but I have no experience. I practiced with pork liver from the butchery, but I failed in doing the slices. The problem was at the slicing step. We have the Mcillwain Tissue Chopper (there will be no other option) and, with it, I just got broken tissue. The liver is too soft and super fragile.
Any suggestion? I am not embedding the tissue in anything. Might that be the answer? Maybe agarose?
You are not clear enough for me to give may be what you want. However, if it is to make slice of thickness not more than 5micrometer, process the liver tissue with paraffin wax and for more than 5micrometer to up to10, use cryostat for frozen sectioning
Hi Vanesa, I have very little experience but what you have sad is probably the right way. I have seen people embedding tissue in agarose before cutting. However I am not sure how much this impact cell viability. I am trying to set up a prep from pig spleen and I will have the same problem. Some people suggested to perfuse the tissue with a gel that would give more stability to the tissue when cutting it, but again I am not sure how this impact the tissue per se.
