Dear fellow researchers,
I am aware of the SRB assay for analyzing viability of cells cultured directly on-to culture dishes. However, my cells are growing on opaque silicon substrates and I want to analyze the cell viability.
Can anyone suggest a modified protocol for analyzing cell viability in this case?

Thanks for the information. I am aware of the staining type you're referring to. We currently had SRB kit available in the lad so I wanted to see if there's anyway I can modify the protocol and use it. Looks like I've got to use the Calcein staining to get the results. Thanks again!
I am not sure but can you detach the cell from the substrate, e.g trypsin, then transfer to another plate before fixation and do the rest.

Thanks for the answer. I can trypsinize them and then perform fixation but several steps in the SRB staining include washing cells with either DI water or 1%acetic acid. To do that, I'll have to centrifuge cells each time. The air drying steps will also get complicated.
