One method is to used thiol additions to DNA which allows DNA to be used as a wire as developed by the Barton lab – see J. Am. Chem. Soc. 2011, 133, 16378–1638 and supporting material. DNA Sequences synthesized by phosphoramidite chemistry to have thiol additions using disulfide phosphoramidites (Glen Research) are used to make DNA modified electrodes. A solution of 50 mM duplex DNA is incubated on a bare gold on mica surface at 4°C overnight, free DNA removed, the surface rinsed, and blocked with1 mM 6-mercapto-1-hexanol, rinsed, and used with proteins for charge transfer measurements.
