To measure the total amount of anthocyanin by the pH differential method, how much of the initial solution of extracted anthocyanin should be added to buffers with different pH (1 and 4.5)?

To 0.5 mL of anthocyanin extract add 3.5 mL of buffer solution pH =1 and read at 510 and 700 nm 15 min later. Repeat the same protocol for the buffer solution pH 4.5.
